A child with a PDH defect
A child has lactic acidosis. Skin fibroblasts from the child and from a healthy control are grown in culture, and pyruvate dehydrogenase (PDH) activity in the child's cells is found to be about 10% of normal. The cells are then given radioactively labelled fuels, and the ¹⁴CO₂ they release and the lactate they secrete are measured.
